Well, what we do is not customizing
I'm not the expert on the subject but (of course) the entire ISO is rebuilt more or less from the bottom up

Developers - not often - they have to much to do to develop Mint - me a lot as you may notice
You are right in your assumption

I know Clem and I use Reconstructor.
It has an option to replace the file and does almost everything automatically for you.
Also the file name we use is splash.pcx

Once you've made your own usplash...
Copy over /usr/lib/usplash/usplash-theme-yoursplash.so (from the chrooted environment) to initrd/usr/lib/usplash/usplash-artwork.so (from the liveCD working root).

Oh.. and we also force usplash into a 1024x768 resolution.. Ubuntu doesn't.. it depends on your artwork I guess. This is done in initrd/etc/usplash.conf

xres=1024
yres=768
