Yes, if you mess up the dd if command, and a lot less likely with the gnome-disks-utility. Back in 2014-ish SanDisk made some USB drives that declared themselves as 'fixed'... It's not being seen as a fixed drive, and doesn't respond to being booted after complete power down, then start up. Thank y...

Not the question you asked, but I will point out there's a better way to accomplish your overall objective. For reasons mentioned in this thread (and others), a persistent drive isn't a good long term choice for a Linux operating system. You would be much better off doing full install to an externa...

It's possible that changes have been made to the original USB flash drive. You might try taking an image of the working flash drive then writing it to the new flash drive. See this thread. When you get to the blank screen, press ctrl+alt+F1 then use sudo nano /etc/fstab. Undo your changes then ctrl+o <enter> to save, and ctrl-x to exit. Well, that saved my bacon!
